I really appreciate the distribution in the data. It is obvious that the variations such as xgirls1cup and 2girlsxcups have the smallest falloff with change in the variable, and then the variations with 0 generally do more poorly than expected if you ignore the context. I think that this is much more than just another "internet meme" comic. It's at least math, even if it doesn't explicitly present an equation or name a theorem.

is there some incredible dislike for 5 girls that even 1 cup couldn't overcome? or the same for 3 cups even though there are 2 girls?

I fail to see how this comic is actually a "joke" in any real way. The idea of searching for variations on girls and cups has potential for amusement, but does not actually deliver anything unexpected. The greatest number of results is for 2 girls, 1 cup, as surmised. So? I assume the "humor" is more like, "hey, this is referring to that 2 girls 1 cup thing! I recognize that reference!" than anything worthwhile.

The alt-text has a joke, though--even if it falls short of an entire 12 Days of Internet Memes thing.

No one claimed it was a joke, but something can still be humorous even if it isn't a joke. Most xkcd don't have "jokes" per se, I think.

The humor resides in
1. doing math(kinda . . statistics?) on something so crude. Is this a type of satire (satire can be treating a serious thing in a nonserious way ("What do we do about the famine? Eat Irish children!") or treating something silly in a serious way (can't think of an example besides this comic))?
and
2. that people would create all those variations from that meme.

Maybe not the best xkcd, but whatever.
