1151: "Tests"

Postby asdfzxc » Mon Dec 24, 2012 5:27 am UTC

Alt-text: In fact, of the boxes is full of Staphylococcus. The wrapping paper is coated in E. coli, though, so it's an understandable mistake. You know, we should really stop accepting gifts from that guy.

This required a Wikipedia visit for me to understand fully.

And yes, we all see the typo.

Better safe than suppurating with bleeding lesions.

Yes, we all see the typo, but do you see the typo you made in the thread subject?

For all the people who are going to be confused: http://en.wikipedia.org/wiki/Gram_staining.

Reminds me of being an undergrad microbiologist, being made to learn the molecular structures of the various proteins in bacterial cell walls in absurd detail. Not fun, especially since all I was interested in was virology.

Now, to test if something is Graham-positive, we need chocolate and marshmallows! ^_^

Oh, that's just great. I got all the way to age 55 before you planted that notion in my brain. I used to like the Super 8! Now what!?

That would include me. Thanks for the link.

I'd like to see someone try to wrap a cube foot of Staphylococcus in E. Coli.

Later, she finds that her presents are gram-everything, and so is her carpet and tree that she's going to spend an hour cleaning.

It wouldn't be that hard, just take a cotton swab, and rub it over the outside of the box after rubbing it in E Coli.

I'm awfully sorry, but this comic doesn't work. She's performing the test in a way that would yield only gram-positive results - she's omitting the decolorization step (clearing the crystal violet/iodine complex with ethanol), so the dark purple ("gram-positive") color remains in all the samples...

I thought this, then decided that the de-colourisation step must be taking place off screen. So it works!

No, when she pours the safranin in panel two, everything is still colored in dark purple. The safranin itself would not have washed out the dark color, just added to its darkness.

Happy now? (I also added a counterstaining step with safranin)
edit: I assumed the second step was iodine. was that the safranin?

I'm assuming that the second panel is supposed to be the safranin counterstain. (Iodine would have been brown). So the microbiologically correct version would look something like this:

She's made another mistake in the procedure: all such tests have to be carried out in low, blue-tinged lighting with funky electronic music playing in the background.

Is is bad that I initially thought she was trying to neutralize any presents that were Artifacts? >.<

Wrapping a present in Murein... now that would be interesting.

And I hope we can all assume that "that guy" is BHG, of course.

What does it mean when Dr. Diaphanous's revised comic presents turn both purple and pink?

At least it's not one of the acid-fast staining techniques... of course, if you don't look for it, you'll never find it. Ignorance = Bliss until you die of leprosy or pulmonary tb.
In the event of pneumothorax, stab me with an 18g angiocath just above the second rib in the midclavicular line.
Thank you.
